Synthetic DNA from gram-negative (GN) or gram-positive (GP) bacteria was spiked in cerebrospinal fluid (CSF) to provide a complex testing matrix, as found (for example) in patients with bacterial meningitis. After extracting the spiked DNA, both assays had a limit of detection of below 10 copies, with excellent specificity and assay linearity. The right panels show specific detection of genomic DNA from a panel of GN or GP bacteria after recovery from spiked CSF.
